The Community star performed three shows at Pharos festival at Auckland's Tapapakanga Regional Park over the weekend, where he surprised the crowd with the world premiere of the film's trailer.
Earlier this year, rumours about the movie swirled as Cuban magazine Vistar revealed photos of Glover and Rihanna on set together.
Grainy fan-footage of the trailer from an Auckland Pharos attendee reveals it will be directed by Hiro Murai, who also directed Glover's seminal music video for 'This Is America'.
Guava Island was billed as 'a Childish Gambino film', and credits Rihanna, Letitia Wright and Nonso Anozie as Glover's co-stars.
The clip begins Glover's character playing guitar alongside his girlfriend, played by Rihanna. In a voiceover, he laments "living in paradise" but not having "the time or means to live here".
The trailer also shows Glover running through a crowded street and being held-up at gun point.
