Police and emergency services were called to the caravan crash just south of Wellsford shortly after 1pm on Monday.
One woman has been seriously injured and is being airlifted to hospital.
Motorists are being asked to avoid the area where possible, as it is expected that the road will remain closed for some time.
Police advise drivers to use State Highway 16 as an alternative route.
